A few pointers that come to mind:

Spell-check it. Read it aloud. Have a friend go over it if you have time. 

Don't go over a page. Keep your paragraphs short. Demonstrate that you were
listening. Emphasize that you're interested in the position. Be careful
about humor--not everyone has a funny bone. 

Send it within 24 hours.
